DESCRIPTION

WARNING

When carbidopa-levodopa is to be given to patients who are being treated with levodopa, levodopa must be discontinued at least eight hours before therapy with Sinemet is started. In order to reduce adverse reactions, it is necessary to individualize therapy. See the WARNINGS and DOSAGE AND ADMINISTRATION sections before initiating therapy.

Carbidopa, an inhibitor of aromatic amino acid decarboxylation, is a white, crystalline compound, slightly soluble in water, with a molecular weight of 244.3. It is designated chemically as (-)-L-alpha-hydrazino-alpha-methyl-beta-(3,4-dihydroxybenzene) propanoic acid monohydrate.

Tablet content is expressed in terms of anhydrous carbidopa, which has a molecular weight of 226.3.

Levodopa, an aromatic amino acid, is a white, crystalline compound, slightly soluble in water with a molecular weight of 197.2. It is designated chemically as (---)-L-alpha-amino-beta-(3,4-dihydroxybenzene) propanoic acid.

Inactive ingredients are cellulose, magnesium stearate and starch. Tablets Sinemet 10-100 and 25-250 also contain FD&C Blue 2. Tablets Sinemet 25-100 also contain D&C Yellow 10 and FD&C Yellow.

Sinemet CR

Sinemet CR (Carbidopa-Levodopa) is a sustained-release combination of carbidopa and levodopa for the treatment of Parkinson's disease and syndrome.

Inactive ingredients in Sinemet CR 50-200 are: D & C Yellow 10, magnesium stearate, iron oxide, and other ingredients. Inactive ingredients in Sinemet CR 25-100 are: magnesium stearate, red ferric oxide, and other ingredients.

The Sinemet CR tablet is a polymeric-based drug delivery system that controls the release of carbidopa and levodopa as it slowly erodes. Sinemet CR 25-100 is available to facilitate titration and as an alternative to the half- tablet of Sinemet CR 50-200.
